Double marker cell line

The invention describes a method for producing a knock in targeted mutation in cells by using a double marker cell line. The double marker cell line genotype is heterozygous, wherein on one allele a promoter is operably linked to a polynucleotide sequence encoding a marker protein and on the other allele a promoter is linked to a polynucleotide sequence encoding another marker protein is present; and obtaining a double marker cell line, especially a double marker embryonic stem (ES) cell line, from this genotype.
